Ph. D thesis at Basrh University to be discussed Investigation of Rhamnolipid Biosurfactant from Pseudomonas aeruginosa with Studying It’s Effect on Biofilm.

Ph. D thesis at the faculty of college of science at the University of Basrah Investigation of Rhamnolipid Biosurfactant from Pseudomonas aeruginosa with Studying It’s Effect on Biofilm, and showed the researcher’s thesis Rhaghad Sharqi Luaibi the isolation and identification of Pseudomonas aeruginosa bacteria from environmental samples and studying the ability of these isolates to produce rhamnolipid biosurfactant  and studying the chemical and genetic identification of rhamnolipids  and studying the biological activity of rhamnolipid as anti-bacterial and anti-biofilm on pathogenic bacteria isolated from diabetic foot infections, as well as studying its effect as antioxidant and anti-cancer agents.

The study aimed to study the biological activity of identified rhamnolipid biosurfactant extracted from Pseudomonas aeruginosa.

The study concluded that Pseudomonas aeruginosa bacteria that were isolated from different regions and sources of Basrah Governorate have the ability to produce Rhamnolipid biosurfactants. Chemical characterization by TLC, FTIR, GC-MS and HPLC indicate the extracted product is rhamnolipid and genetic diagnosis by rhlAB gene confirmed the chemical identification of rhamnolipid. Rhamnolipid extracts exhibit antibacterial activity, anti-biofilm activity, anti-adhesive activity, disruption preformed biofilm activity  against pathogenic bacteria isolated from diabetic foot infection. Rhamnolipid exhibited synergistic effect with two antibiotics Gentamicin and Ciprofloxacin by increasing the inhibition zone diameter, and rhamnolipid showed good antioxidant and had an inhibitory effect on MCF7 and SKG cell lines.

The study recommended the use of other media and inexpensive compounds in the production of rhamnolipid, studying anti-fungal activity and antiviral activity, and using genetic engineering to enhancement the production of rhamnolipid and testing the synergistic effect with more antibiotics.
